Proposed Working Mechanism of Myostatin
Myostatin binds to activin type II receptors (ActRIIA and ActRIIB) on muscle cells, which then recruit and activate type I receptors to initiate intracellular signaling. It further triggers a cascade of phosphorylation events inside the cell, as observed in in vitro studies.
This signaling cascade activates SMAD proteins, specifically SMAD2 and SMAD3, which combine with SMAD4 and move into the nucleus of the cell.
Once inside the nucleus, the SMAD complex regulates gene expression. It suppresses genes involved in muscle cell growth and differentiation while promoting genes that inhibit muscle development in preclinical investigational models. This signaling has been associated with reduced proliferation and differentiation of muscle precursor cells in preclinical studies.
